I was experiencing really slow startup times for iTunes, something like 23 bounces in the dock. I have 2500 songs. I tried converting my smart playlists to regular ones and it didn't help. I also repaired disk permissions with Disk Utility and tried clearing cache files of other programs.

I have 2 identical hard drives, one of which runs as a backup using Carbon Copy Cloner. I ran Norton Disk Doctor and Speed Disk on the backup drive and booted into that drive. Now iTunes comes up in 2 bounces.

So if converting smart playlists doesn't work for you, try defragmenting your drive. I also highly recommend having 2 identical drives for avoiding the click of death and being able to easily run things like Norton without having to boot from a CD.
