Yep, that's why the trick was created in the first place. In the early days of the Mac (the original 128K Mac and the 512K, probably even the Plus days), it was not uncommon to have a floppy that contained your OS and the app you wanted to run. If you wanted to boot off of a different floppy than the one in the drive, just hold down the mouse button as you boot. You'd then see the flashing question mark, and pop in your new floppy!
