Yes, you can accomplish this by using an AppleScript.

Then, to meet your one-click requirement, put a link to this script in your dock, or go to the mouse preference pane and assign the script to one of your mouse buttons, like the side button (button 4) on the Mighty Mouse, for example.

If you look at the script in the Script Editor, it is a very simple script. It just reads the current state of the hot corners in and then either disables them or sets them back to what they were depending on whether the hot corners are disabled or enabled. It then kills the dock, which is then reloaded with the new hot corner settings. It all happens very quickly.

I've put this in my applications folder as I don't need frequent access to it, but it really comes in handy as there are some apps, mostly games, that I use that I find myself accidentally activating hot corners.
