10.4: Disabling Spotlight in target disk mode System
I recently connected my new MacBook to my iMac with the MacBook in target disk mode and switched off Spotlight on the mounted disk (i.e. the MacBook's hard drive) via the System Preferences -> Spotlight -> Privacy panel.

It turns out this disables Spotlight for that drive on the MacBook, even when it is booted up normally and is no longer being used as a FireWire drive. It then has to be enabled again (and the index recreated) in the System Preferences on the computer that was used in target disk mode.

This makes sense, as Spotlight works on a per-drive basis (i.e. the index for a given drive is stored on that drive, no matter where or how that drive is mounted). If you want to turn off Spotlight on a drive without having to completely re-index it later, you can use the command line:
sudo mdutil -i off "Path_To_Volume"
This will turn off indexing but preserve the actual index files. When you're ready to turn Spotlight back on, use:
sudo mdutil -i on "Path_To_Volume"
Spotlight will be re-enabled for the volume, and will only need to index newly added files. All this applies to firewire drives and partitions as well, by the way.
