I haven't found a way to reliably test time this, but it's perceputally noticeable, and several friends who tried it confirmed it:

Safari seems to slow down as the number of items in the Bookmarks bar and Bookmarks menu grows. This slowdown appears to be more noticeable when launching Safari. So to speed it up a bit, try to move as many bookmarks as possible out of the Bookmarks bar and menu, and into other collections.

[robg adds: This is the latest in a continuing series of Safari speed-up tips. To summarize what we have so far, you can make Safari much faster by disabling JavaScript and RSS, disabling unused languages and deleting autofill entries, removing cached FavIcons, and using short names for bookmarks.]
