In the bottom left corner of a QuickTime window, there is a slider for the volume. If you click on the speaker icon, it will mute the volume and disable the slider control -- but leave the slider in its current position. Click on the icon again to "unmute" the volume and allow control of the slider.

The only problem with using the mute function is that putting QuickTime Player in the background and then bringing it to the foreground again will unmute the movie. Strange behavior.

Doesn't work for my system! The mute button on my keyboard works fine, but clicking the icon on the player doesn't do anything but show the slider. I have an iMac G3 450 MGz, OSX 10.3.9, QT 7.02.
